When I was first making brushes, I'd hold the slab in one hand and drill with the other. It wasn't easy, but it got the job done. Patrick suggested I use the vise. I tried it with sponges around the sides to keep the clay from being marked, but it was too soft. The clay bulged out from pressure distortion. So I asked if he could find me two boards in his giant pile of random wood pieces, and lo and behold! They can screw to the vise and hold the clay snug between them, and the stability of the wood creates opposite pressure, ensuring the slab remains flat on either side.
